ELISA实验操作较繁杂，操作不当将引起较大的误差。小鼠E钙粘着蛋白/上皮性钙黏附蛋白酶免试剂盒，（E-Cad）ELISA检测试剂盒在操作中应注意以下5个方面。
1．随着病情的进展或由其他因素影响，某些抗体在机体内的含量发生大幅波动，因此有必要对标本进行预处理，例如对血清标本作适当稀释，或离心分离血脂等。间接法测定中，标本适当稀释还可降低非特异性反应。
2．加样一般使用加液器，在ELISA中一般有4次加样：加标本、加酶结合物、加底物和加终止液。加标本时必须每份标本使用一支移液器吸嘴，以免交叉污染。加酶结合物、底物和终止液时则不需更换吸嘴。
3．在成批手工检测中，还应注意操作时差的影响。加样及混匀时间的差异，使抗原抗体反应和酶促反应时间不*，对竞争法及定量检测影响很大，不注意可能会导致错误结果或定量不准。
4．洗涤是ELISA操作过程中决定实验成败的一个关键步骤。目的是除去未结合的免疫反应物，终止抗原抗体反应，除去标本中与反应无关的成分、游离的酶标物以及反应过程中吸附于固相载体上的非特异性物质?？捎孟窗寤窗寤蚴止は窗澹罢呦吹又柿拷虾?，各反应孔洗涤效果均一，批内、批间差异小，手工洗板应注意控制各孔洗涤效果，差异不宜太大。
5．准确判定结果须使用ELISA测读仪（酶标仪），比色法判读可选择单波长或双波长，根据反应液颜色的不同选用不同波长的滤光片，以空白孔校零测定反应孔的吸光度值（A值）。酶标仪具有良好的重复性。
